The steps to get a game running might vary widely per game. A good place to start in such cases would be, like I found for you, a third-party users guide.

Attempting to run the DK2 file directly as an admin I already tried and that still comes up with the inaccessible error. If you have the space, can you try installing to C as a test? I suspect it won't be a factor, but best to rule it out. If this has no effect, can you try restarting your machine in Safe Mode with Networking your desktop will not look pretty and see if this has any effect on the download process? Running in safe mode got the game to attempt to start, but all it did was open into a black screen and crash before loading anything.

Clicking repair installation in safe mode didn't give me the 'download failed' message but didn't fix anything. Now I'm back in normal mode and it's back to not doing anything when opened from origin and saying I don't have appropriate permissions when opened from the start menu.

One other thing, before, the game icon was the kind of icon you get for an unknown exe file. Now after opening it in safe mode the program does have the proper DK2 icon. If that means anything. From the evidence given I strongly suspect that your security software is blocking the game. Safe Mode will have zero 3rd party programs including security software running which could interfere, and the game seemed to start.

It failed and went to a black screen when it started, but that's perfectly normal as in Safe Mode your video card won't be active part of why it looks so hideous.
